IgA Nephropathy (IgAN)

IgA Nephropathy (IgAN), also known as Berger’s Disease, is an autoantibody-mediated kidney disease. For people living with IgAN, the body makes an abnormal form of a protein called immunoglobulin A (IgA). While IgA is a normal part of the immune system, in people with IgAN the body’s immune system forms autoantibodies against the abnormal form. These autoantibodies and the immunoglobulin proteins form immune complexes, which build up in the glomeruli—the tiny filters in the kidney—causing inflammation and scarring. This impairs how waste is filtered in the kidneys and can lead to chronic kidney disease and kidney failure.

IgAN symptoms include blood in the urine, protein in the urine, high blood pressure, and swelling (edema).

The management of IgAN is primarily focused on reducing proteinuria as this is correlated to improved outcomes and reduced risk of kidney failure.

Unmet need

IgAN is the most common primary glomerular disease worldwide. The unmet need in IgAN is high and there is a well understood need for treatments that would prevent the production of abnormal IgA and associated autoantibodies. Because many patients with IgAN are asymptomatic or have minimal urinary abnormalities, the disease is likely underdiagnosed, and its true prevalence may be higher than reported.

How CLYM116 may be able to help

APRIL signaling promotes survival of plasma cells, the source of many autoantibodies. It also regulates the process in which some B cells begin to express IgA. In IgAN, blocking APRIL signaling may prevent the production of abnormal IgA and the resulting autoantibody production and immune complex formation that leads to kidney damage.

Through its anti-APRIL mechanism of action, CLYM116 has the potential to reduce IgA and be a disease-modifying treatment for IgAN. CLYM116 is currently in preclinical development, with an IND or CTA filing planned by the end of 2025.
